The Portuguese sat out the 3-2 win at Rayo Vallecano on Saturday, but Zinedine Zidane said both at the weekend and in Monday's press conference that Ronaldo, 31, would recover in time.
But Anton Meana of Radio Marca has reported that the former Manchester United man is not in the starting lineup to face City at the Etihad tonight.
With the ten-time European Cup winners also sweating over the fitness of Karim Benzema, Zidane could be missing two of his most potent weapons for his most important game as a manager.
